Abstract:

When tobacco is smoked, chewed or snuffed, nicotine is absorbed by the lungs or mucous membrane and quickly moved into the bloodstream, where it is circulated throughout the brain. In fact nicotine is highly dangerous to be consumed in any form. The present study was conducted to know the adverse effects of nicotine on the platelet aggregation in cerebral microvessels of mice. Male mice of average weight 33 g were injected with Saline (control) or Nicotine (1 mg/kg, 0.1 ml/10 g) one hour before the experiment. Animals were anaesthetized and trachea was intubated. Craniotomy was performed and a window was opened on the skull. Layer of dura was removed. Brain surface microvessels were exposed and animal was placed on the microscope stage. Microscope was connected to a monitor and VCR to record all events. Exposed brain surface was continuously irrigated with ACSF solution. After the body temp was maintained at 37? C, Sodium fluorescein (2%, 0.1 ml/10 g) was injected i.v. through tail injection. High intensity mercury light was switched on to induce photochemically induced platelet aggregation. Appearance of the 1st platelet aggregation and total blood flow stop were timed in seconds. Our results showed that in the animals treated with nicotine, venule did not show any alteration in the platelet aggregation timings in comparison to the controls. But in arterioles platelet aggregation timings were significantly accelerated (P

Antioxidants alleviate nicotine-induced platelet aggregation in cerebral arterioles of mice in vivo.

Experimental data on the effect of nicotine on cerebral microvessel thrombosis is lacking. Therefore, this study was carried out to elucidate the effects of nicotine on platelet aggregation in cerebral (pial) microcirculation of the mouse, and the possible protective effect of vitamins C and E. Male TO mice were divided into six groups, and injected i.p. with saline as a control, nicotine (1 mg...

Platelet Aggregation in Mouse Cerebral Arterioles

Background and Purpose: Epoxygenase metabolites of arachidonic acid are produced by several tissues and have been shown to inhibit in vitro platelet aggregation. The purpose of the present investigation was to determine whether 14,15or 8,9-epoxyeicosatrienoic acid, epoxygenase derivatives of arachidonic acid, affect the speed of platelet aggregation in in vivo mouse cerebral arterioles. Methods...

Angiotensin delays platelet aggregation after injury of cerebral arterioles.

Endothelium of cerebral surface vessels (pial arterioles and venules) was injured with a light/dye technique in anesthetized mice. This induced platelet aggregation at the site of injury. The onset of aggregation was monitored through a microscope in mice given angiotensin II acetate, 4 micrograms i.v. 30 minutes earlier. Aggregation latency was compared with that in vehicle treated (saline) mi...

Sub-acute Exposure to Benzene Accelerates the Aging Process of Red Blood Cells; an In vivo Study

Background: The well-known toxic effects of benzene toxicity are bone marrow depression, reduction in blood cell counts, and induction of leukemia and aplastic anemia. This study was designed to evaluate biomarkers of aging in red blood cells (RBCs). Methods: Mice were exposed to benzene (50, 100, and 200 mg/kg/day) orally for 28 days. A group of benzene-exposed mice were injected intraperiton...

Shear-induced platelet aggregation in cerebral ischemia.

BACKGROUND AND PURPOSE Recent evidence has suggested that shear-induced platelet aggregation is an important mechanism of thrombosis at arterial bifurcations or stenoses. We measured shear-induced platelet aggregation with a new apparatus in patients with cerebral ischemia and also studied correlations with other hemostatic parameters as well as the effect of antiplatelet agents. METHODS The ...
